چکیده

Michael Shea and Lewis J. Kleinsmith Department of Zoology, University of Michigan, Ann Arbor, Michigan 48104 Received December 1,1972 SUMMARY Phosphorylated non-histone chromatin proteins are shown to stimulate the synthesis of RNA in a cell-free system using rat liver RNA polymerase and rat DNA as template, This stimulation is not observed when DNA's of other species are used as template, or when the phosphorylated proteins have been treated with alkaline phosphatase.

Study of the loosely bound non-histone chromatin proteins. Stimulation of deoxyribonucleic acid-templated ribonucleic acid synthesis by a specific deoxyribonucleic acid-binding phosphoprotein fraction.

The loosely bound chromatin proteins of Ehrlich ascites hyperdiploid cells have been prepared by extraction of chromatin with 0.35 M NaCl. Sodium dodecyl sulfate gel electrophoresis of the 0.35 M NaCl-soluble chromatin proteins reveals high heterogeneity with a molecular weight range of 10,000 to 170,000. The 0.35 M NaCl-soluble chromatin proteins contain many components similar to the more tig...

Phosphorylation of non-histone proteins in the regulation of chromosome structure and function.

Non-histone chromosomal proteins are phosphorylated and dephosphorylated within the intact nucleus by two independent sets of reactions, a protein kinase reaction which transfers the terminal phosphate group of a variety of nucleoside and deoxynucleoside triphosphates to serine and threonine residues in the proteins, and a phosphatase reaction which cleaves these phosphoserine and phosphothreon...

Myocardial chromatin activation in experimental hyperthyroidism in rats. Role of nuclear non-histone proteins.

Experimental hyperthyroidism induced in rats by daily injections of 20/*g of 3',3',5-triiodothyronlne (T,) resulted in a prompt increase in cardiac weight and RNA content. The mechanism of the RNA synthesis stimulation was studied by comparing template activity of myocardial chromatin from rats treated with T3 for 7 days with chromatin from euthyroid controls. Hyperthyroidism resulted in signif...

Control of RNA synthesis by chromatin proteins.

The effect of chromatin proteins on template activity has been studied. Using both E. coli RNA polymerase and calf thymmus polymerase B we have measured the number of initiation sites on chromatin and various histone-DNA complexes. Chromatin can be reconstituted with histone proteins alone and this complex is still a restricted template for RNA synthesis. The removal of histone f1 causes a larg...

Non-histone-protein phosphorylation in rat salivary glands after treatment with isoproterenol, dibutyryladenosine cyclic monophosphate and dibutyrylguanosine cyclic monophosphate.

A single injection of isoproterenol to rats produces a wave of DNA synthesis that reaches a peak 28 hlater (Barka, 1965; Baserga, 1966). This system has become increasingly useful as a model for cell proliferation in viuo; one of the first steps required to stimulate resting cells into cycle is thought to be gene activation (Lieberman, 1970; Baserga & Stein, 1971). Recent evidence suggests that...
